According to Official Guinness Records,

The fastest swim long course 100 metres backstroke by a male is 51.85 seconds, achieved by Ryan Murphy (USA) during the final of the men’s 4 x 100 m medley relay at the 2016 Olympic Games, in Rio de Janeiro, Brazil, on 13 August 2016.Overall at the Rio Olympics Ryan won three gold medals in the 100 m and 200 m backstroke and the 4 x 100 m medley.
